Sa fille encore enfant, la mère de Brenda Blethyn a dit un jour d'elle qu'elle allait devenir actrice. Pourtant, ce n'est qu'à 27 ans que Brenda prend ses premiers cours de théâtre.
En 1980, elle débute à la télévision sous la direction de Mike Leigh.
Au début des années 1990, elle tourne ses deux premiers films de cinéma aux Etats-Unis (Les Sorcières, de Nicolas Roeg et Et au milieu coule une rivière, de Robert Redford). En 1996, et alors que c'est seulement son troisième film de cinéma, elle remporte le prix d'interpréation au festival de Cannes pour sa participation à Secrets and Lies de Mike Leigh.
